Samenvatting

Het ontrafelen van de mechanismen van veroudering vormt een van de grote uitdagingen in de biologie. In de laatste eeuw zijn uiteenlopende verouderingstheorieën naar voren gebracht. De genen die defect zijn bij progeroïde syndromen bij de mens kunnen in laboratoriummuizen worden uitgeschakeld door genetische modificatie. Vooral uit het onderzoek van muizen met defecte DNA-reparatiegenen is gebleken dat ophoping van endogene DNA-schade een belangrijke rol speelt bij het verouderingsproces. Het optreden van deze schades als gevolg van de stofwisseling veroorzaakt op zijn beurt een overschakeling van een stofwisselingsmodus die gericht is op verbranding en groei naar een modus die meer gericht is op bescherming en conservering.
Ouder worden gaat gepaard met verliezen op lichamelijk, functioneel, cognitief en sociaal vlak. Toch blijven de meeste ouderen zich lang goed voelen. Hoe slagen ze daarin? Op zoek naar een antwoord hebben gerontologen welbevinden in de tweede levenshelft in verband gebracht met veranderingen in de hersenactiviteit, het cognitief functioneren, de motivatie, de sociale context en de persoonlijkheid.
Dé oudere bestaat niet; er zijn grote verschillen in de wijze waarop mensen oud worden en door de sociale omgeving oud worden gemaakt. Verschillende theorieën die een verklaring willen geven voor dat proces, worden besproken; ze belichten elk een ander aspect. Ook wordt een aantal belangrijke wetenschappelijke stromingen in de gerontologie beschreven en wordt aan de hand van een voorbeeld uitgelegd hoe die inzichten kunnen worden toepast bij het begrijpen van gerontologische vraagstukken.
