Samenvatting

De genetische epidemiologie zoekt naar variatie in het humane genoom als determinant van ziekte. De speciale vorm van epidemiologie kent zijn eigen onderzoeksdesigns en analysemethoden. Het hoofdstuk begint met een korte en algemene introductie van de genetica, waarna het familie- en het tweelingonderzoek worden besproken. Hierbij komen de principes van koppelingsanalyse en recombinatie (inclusief LOD-score) aan bod. Daarna wordt het genetisch associatieonderzoek uitgelegd, waarin begrippen zoals linkage disequilibrium, haplotypeanalyse, TDT-analyse, genoomwijd onderzoek en populatiestratificatie de revue passeren. Het hoofdstuk laat zien hoe met behulp van mendeliaanse randomisatie een gerandomiseerde trial kan worden nagebootst en geeft voorbeelden van Nederlands biobankonderzoek.
