Genetica van aandacht en aandachtsproblemen. Kan executief functioneren dienen als cognitief endofenotype?

Abstract

Aandachtsproblemen en Attention Deficit/Hyperactivity Disorder ( adhd ) zijn sterk genetisch bepaald. Dit geldt voor zowel jongens als meisjes, en gedurende de hele kindertijd. De zoektocht naar genen voor een complexe stoornis als adhd blijkt echter lastig. Een mogelijke oorzaak hiervoor is dat adhd een heterogeen fenotype laat zien, zowel cognitief als gedragsmatig. Tevens wordt aangenomen dat verschillende, met elkaar interacterende genen, verantwoordelijk zijn voor de stoornis. Om meer zicht op het genetische en daarmee biologische mechanisme te verkrijgen dat adhd veroorzaakt, wordt onderzoek gedaan naar gerelateerde, onderliggende kenmerken van de stoornis, zogenaamde endofenotypes. Deze zouden een deel van de complexe genetische puzzel, die adhd veroorzaakt, kunnen blootleggen. Executieve functies leken veelbelovende cognitieve endofenotypes. Na een decennium van onderzoek lijken zij echter vooralsnog geen short cut te zijn naar de genen van adhd.
