Functional studies on c.1347C>T, a polymorphism modulating phenotypic outcome in X-linked adrenoleukodystrophy

We have recently shown that the minor allele of the CYP4F2 polymorphism c.1347C>T increases the risk of developing the most severe phenotype of X-linked adrenoleukodystrophy. Using cellular models of VLCFA accumulation, we have shown that p.V433M, the amino acid substitution encoded by c.1347C>T, reduces the omega-oxidation of VLCFA to very long-chain dicarboxylic acids. This effect was not mediated by decreased catalytic activity of CYP4F2 but by a marked reduction in CYP4F2 protein levels, despite unaltered levels of CYP4F2 mRNA. Our current work focuses on resolving the deleterious effect of p.V433M on CYP4F2 protein levels. …
