Augmentatie van antidepressiva: mogelijkheden voor verbeterde werkzaamheid?

Ondanks dat techieken als neuro-imaging (mri en pet) maar ook dna- en eiwitonderzoek (genomics en proteomics) bijdragen aan verbeterde inzichten, is de huidige kennis over de psychopathologie van affectieve stoornissen nog steeds beperkt. Het ziektebeeld depressie is zo divers dat het onwaarschijnlijk lijkt dat er slechts een enkel farmacologisch mechanisme bij betrokken is. Dit maakt de speurtocht naar nieuwe geneesmiddelen aanzienlijk lastiger. Tot op heden is er dan ook sinds de ontwikkeling van de monoamine oxidaseremmers (maoi) in de jaren zeventig van de vorige eeuw geen wezenlijke farmacotherapeutische vernieuwing geweest op dit terrein. …
